Quickly optimise flask nginx file uploads/form request with file in it (FLASK/AWS/NGINX/GUNICORN)

I host an app that deals with very big file uploads that are temporarily stored in an EC2 instance on AWS then sent off to S3, all of that works fast but for some reason the actual request takes well over a minute to fire up before doing anything. I have messed a bit with the config but don't know enough, I have heard there are ways to optimise it but don't want to break it.
